Abstract

Ulcerativecolitis(UC)isachronicrefractorynon-specificintestinalinflammatorydiseasethatisdifficulttobecured.Thediscoveryofnewulcerativecolitis-relatedmetabolitebiomarkersmayhelpfurtherunderstandUCandfacilitateearlydiagnosis.ItmayalsoprovideabasisforexplainingthemechanismofdrugactioninthetreatmentofUC.CompoundSophoraeDecoction(CSD)isanempiricalformulausedintheclinicaltreatmentofUC.Althoughitisknowntobeefficacious,itsmechanismofactioninthetreatmentofUCisunclear.ThepurposeofthisstudywastoinvestigatethechangesinendogenoussubstancesinUCratsandtheeffectsofCSDonmetabolicpathwaysusingthemetabonomicsapproach.MetabolomicsstudiesinratswithUCandnormalratswereperformedusingLC-MS/MS.RatswithUCinducedusingTNBSenemawereusedasthestudymodels.Metabolicprofilingandpathwayanalysisofbiomarkerswasperformedusingstatisticalandpathwayenrichmentanalyses.36screenedpotentialbiomarkerswerefoundtobesignificantlydifferentbetweentheUCandthenormalgroups;itwasalsofoundthatCSDcouldmodulatethelevelsofthesepotentialbiomarkers.CSDwasfoundtobeefficaciousinUCbyregulatingmultiplemetabolicpathways.
